摘要 本文利用钴原卟啉对H_2O_2氧化有很高催化活性的特点,以钴原卟啉修饰玻碳电极为基底,戊二醛交联法固定胆固醇氧化酶,以电聚合邻苯二胺膜抗干扰、抗毒化.得到了线性范围为0.1~2mmol/L,响应时间在30s以内,可连续使用150次的胆固醇酶电极. A new amperometric sensor for free cholesterol has been fabricated based on a high electrocatalytic activity of cobalt protoporphyrin to the oxidation of H2O2. A layer of cobalt pro-toporphyrin was absorbed on the glassy carbon electrode, the enzyme film consisting of bovine serum albumin and cholesterol oxidase was put on it, then a layer of poly(ο-phenylenediamine) film was electropolymerized out of the enzyme film to avoid interference and fouling. The linear range of concentration was 0. 1 mmol/L -2 mmol/L. The response time was less than 30 s. The electrode was used for 150 times assays without loss of any enzymatic activity.
