产生廿碳五烯酸等多不饱和脂肪酸真菌的研究Ⅲ.SM96菌株工业发酵条件 被引量:1

摘要 为考察SM96菌株利用紫苏油发酵生产Ara(花生四烯酸)和EPA(廿碳五烯酸)过程中需要控制的几个基本参数,利用气相色谱法检测不同发酵条件下,菌株SM96产Ara和EPA情况;并通过正交实验优化发酵培养基配方,结果表明,有利于SM96菌株产Ara和EPA的条件为接种量体积分数4%、培养温度25℃、初始pH为6 0、装液量20%;优化的发酵培养基配方葡萄糖为25g·L-1、蛋白胨为5g·L-1、紫苏油为35g·L-1、酵母膏为2g·L-1。 Attempt has been made to find several basic parameters which should be controlled in the process of converting perilla oil to an oil containing Ara and EPA by SM96.Under various fermentation conditions, contents of EPA and Ara in mycelia of SM96 were measured by gas chromatography. Fermentation medium for mulae were optimized by orthogonal design. The optimum conditions for Mortierella sp. to produce Ara and EPA were 4% of seed volume;25℃ of fermentation temperature;6.0 of initial pH;20% of medium volume;optimal medium compositions including glucose 25(g·L^(-1)), peptone 5g·L^(-1), perilla oil 35g·L^(-1) and yeast extract 2g·L^(-1).
