基因工程双特异性抗体构建模式研究进展 被引量:2

摘要 为了增强抗体的效应功能 ,人们尝试多种方法改造抗体分子 ,双特异性抗体是改善抗体治疗效果的发展方向之一 ,现已成为抗体工程研究领域的热点。随着分子生物学技术的迅速发展 ,出现了基因工程双特异性抗体的多种构建模式 ,并且有多种基因工程双特异性抗体制剂已经用于肿瘤的临床诊断和治疗试验。本文对基因工程双特异性抗体的构建模式按照双特异性微抗体、双链抗体、单链双特异性抗体和多价双特异性抗体 4类进行了综述。 In order to improve the therapeutic utility of antibodies, different modifications have been suggested. One of them concerns the generation of bispecific antibody (BsAb). BsAbs are artificially created antibodies with two different specific antigen-binding sites, which have a significant potential for cancer therapy. Genetic engineering has led to the development of various BsAb formats, with the aim of optimizing their structure, minimizing toxicity. Here, different formats for BsAbs are summarized and discussed.
