一种新的凋亡抑制蛋白Livin在非小细胞肺癌组织中的表达

Expression of Livin,a novel inhibitor of apoptosis protein family member,in tissues of non-small cell lung cancer

目的 探讨一种新的凋亡抑制蛋白 (inhibitorofapoptosisprotein ,IAP)Livin两种异构体Livinα和Livinβ在非小细胞肺癌 (NSCLC)组织中的表达及其与NSCLC组织类型、放化治疗的关系。方法 采用逆转录多聚酶链式反应 (RT PCR)检测NSCLC组织中Livinα和LivinβmRNA的表达 ,并用westernblot分析Livin蛋白的表达。 结果 4 8例NSCLC组织中Livin表达阳性率为 2 2 .9% ,其表达与NSCLC组织学类型相关 ,腺癌中Livin表达阳性率达 4 5 .5 % ,明显高于鳞癌和大细胞癌 (P <0 .0 1 ) ,而癌旁组织和良性疾病肺组织未检出阳性结果 ;Livin表达还与肿瘤治疗相关 ,放化治疗可明显诱导Livin表达 (P <0 .0 1 )。结论 Livin异构体在非小细胞肺癌组织中表达 ,作为肺腺癌的分子标志物 。

Objective To study the expression of two isoforms(i.e.Livinα & Livinβ) of Livin,a novel inhibitor of apoptosis protein(IAP)family member,in non small cell lung cancer(NSCLC) and to explore relationship between Livin expression and NSCLC histological types,and changes of its expression after radio and/or chemotherapy,respectively.Methods Livin mRNA and protein expression levels in NSCLC tissues were detected by reverse transcription polymerase chain reaction(RT PCR) and western blot assay,respectively.Results Livin mRNA and protein were expressed in 11 of 48 NSCLC cases(22.9%).The positive rate of Livin expression in lung adenocarcinoma reached 45.5%,much more than that of lung squamocellular carcinoma and large cell carcinoma( P <0.01).No Livin expression was detected in para cancerous or benign disease lung tissues.Radiotherapy and/or chemotherapy induced gene expression of Livin remarkably.Conclusion Livin isoform expression was detected in NSCLC.As molecular marker of lung adenocarcinoma,Livin may be served as a new target for apoptosis inducing therapy of NSCLC.
