诱导型一氧化氮合酶在脊髓损伤后组织中的定位和定量研究 被引量:1

摘要 目的 研究诱导型一氧化氮合酶 (Induciblenitricoxidesynthase ,iNOS)在大鼠脊髓损伤后局部分布和表达的规律。方法 分别于大鼠脊髓压迫伤后 6、12、2 4、4 8、96h等 5个时间点 ,应用免疫组织化学和图像分析技术 ,对脊髓组织中iNOS的分布和表达规律进行定位和定量研究。结果 iNOS在正常对照组织中没有表达 ,损伤后开始出现 ,免疫组化阳性部位为细胞浆内的棕色染色部分 ,主要分布在脊髓的前角和中央管周围的大多角形细胞以及后角的小圆细胞 ,图像分析表明伤后 12、2 4、4 8h等组的测量值分别与正常对照组相比有显著性差异 (P <0 .0 5 )。结论 脊髓损伤后脊髓组织中存在iNOS表达 。 Objective To study the distribution and expression of inducible nitric oxide synthase(iNOS) in spinal cord injury.Methods The spinal cords of SD rats were compressed by the formal methods.6,12,24,48 and 96 hours after the injury,immunohistochemical and image analysis methods were used to study the the distribution and expression of inducible nitric oxide synthases in spinal cord tissues.The spinal cord tissues of normal rats were used as controls.Results The immunohistochemically positive areas were located in brown-stained part of cytoplasm in cells of anterior horn , central canal and posterior horn . iNOS were expressed in all injured groups but not in normal control group.There were significant differences between 12-48 hours after-injury groups and the normal control group(P<0.05).Conclusion There are expressions of iNOS in spinal cord tissues of rats with spinal cord injury. This fact would be useful for studying the roles of nitric oxide in spinal cord injury.
