摘要
目的 探讨金雀异黄素对MCF乳腺癌细胞抑制作用的机理。方法 在体外培养的MCF乳腺癌细胞中加入不同浓度的金雀异黄素 ,2 4h后收集细胞和培养液 ,分别采用酶法和分光光度法测定了细胞匀浆液和培养液中一氧化氮合酶 (NOS)、谷胱甘肽过氧化物酶 (GSH Px)活性及一氧化氮 (NO)、还原性谷胱甘肽(GSH)及丙二醛 (MDA)含量。结果 加入金雀异黄素作用后 ,细胞匀浆液和培养液中NOS、GSH PX活性及NO含量明显升高 (P <0 0 5 ,P <0 0 1 )。GSH、MDA含量则明显降低 (P <0 0 5 ,P <0 0 1 )。结论 金雀异黄素可影响MCF乳腺癌细胞NO生成系统和抗氧化系统 。
Objective To investigate the mechanism that the genistein inhibit the MCF human breast cancer cell. Methods The activities of glutathione peroxidase(GSH Px), nitric oxide synthetase(NOS) and the contents of nitric oxide(NO), glutathione(GSH), malondialdehyde (MDA)both in culture supernatant and cells were examined by enzyme method and spectrophotometry respectively. Results\ With genistein increasing, the activities of GSH Px, NOS and the contents of NO increased significantly( P <0 05), while the contents of GSH, MDA decreased both in culture supernatant and cells( P <0 05). Conclusion\ Genistein can affect NO synthesis and antioxidation in MCF breast cancer cells. It may be one of the mechanisms that the gnistein inhibit the MCF breast cancer cells growth.
