摘要
目的 探讨治疗药物对帕金森病患者外周血单核细胞 (PBMC)分泌白细胞介素 6(IL 6 )和肿瘤坏死因子 α(TNF α)的影响。 方法 用酶联免疫吸附方法检测健康对照组、培高利特+美多芭组和美多芭组共 2 5例受试者的外周血单核细胞 ,分别在无刺激、含刀豆蛋白A (ConA )或含培高利特的不同培养方式下 ,分泌IL 6和TNF α的浓度。 结果 IL 6分泌水平 :在ConA刺激下 ,培高利特 +美多芭组和美多芭组均显著低于对照组 (P <0 0 5 ,P <0 0 1) ,且培高利特 +美多芭组显著高于美多芭组 (P <0 0 5 ) ;培高利特刺激下 ,3组差异无显著性 (P >0 0 5 )。TNF α浓度 :培高利特 +美多芭组在ConA刺激下较无刺激和培高利特刺激均显著增高 (P <0 0 1) ;且不同培养基培养后 ,3组间比较差异均无显著性 (均为P >0 0 5 )。 结论 药物治疗的PD患者PBMC分泌IL 6的能力显著低于健康对照者。培高利特可能通过改善和恢复帕金森病患者PBMC分泌IL 6能力发挥治疗作用 ,同时不增加TNF α的分泌 。
Objective To investigate the effects of anti-parkinsonian drugs on cytokines(IL-6 and TNF-α) produced by PBMC in PD patients. Methods 25 subjects were divided into 3 groups : normal controls(NC),pergolide and Madopar treated PD patients(P+M group),Madopar treated PD patients(M group). PBMC were cultured in the presence of pure RPMI-1640,RPMI-1640 with ConA and RPMI-1640 with pergolide,respectively. The amount of IL-6 and TNF-α in the culture media was measured by enzyme-linked immunosorbent assay (ELISA). Results IL-6 production amount by PBMC when stimulated with ConA was significantly lower in group P+M and M than in group NC( P <0.05 and P <0.01),while the amount in group P+M was significantly higher than in group M ( P <0.05);when stimulated with pergolide there were no significantly differences among group P+M,group M and NC ( P >0.05). In group P+M,the concentration of TNF-α was significantly higher in the presence of ConA than in the presence of pure RPMI-1640 and in the presence of pergolide( P <0.05);With the different culture media,there were no significantly differences of TNF-α production by PBMC among group NC,P+M and M( P >0.05). Conclusions The ability of amount by PBMC of PD elderly patients to excrete IL-6 was significantly decreased in the presence of ConA. Pergolide might arise the excretion of IL-6 by PBMC during the treatment process of PD and at the same time does not arise the excretion of TNF-α,which might be neurotoxic.
