摘要
为观察痹康灵对骨关节炎 (OA)的治疗效果并探讨其作用机制 ,将 32只中国白兔分为正常对照组 (A组 )、空白对照组 (B组 )、西药对照组 (C组 )、中药治疗组 (D组 ) ,每组各 8只 ,A组不造模 ,B、C、D组均采用Hulth法建立兔OA模型。术后 3天给予A和B组生理盐水 ,C组罗非昔布溶液 ,D组痹康灵 ,每天各 6ml·kg-1,灌胃给药。自术后第 1周每天驱赶动物 2次 ,共 30分钟 ,4周后取右侧胫骨平台关节软骨和滑膜进行光镜观察。采用免疫组化检测法分别检测关节软骨和滑膜中的白细胞介素 1β(IL β)、基质金属蛋白酶 1(MMP 1)、基质金属蛋白酶 3(MMP 3)的水平。结果显示 ,B组IL 1β ,MMP 1、MMP 3的水平均非常显著高于A组(P <0 .0 1) ,并均显著高于C组 (0 .0 1<P <0 .0 5 ) ;B组IL 1β、MMP 3水平显著高于D组 (0 .0 1<P <0 .0 5 ) ,而MMP 1水平则非常显著高于D组 (P <0 .0 1) ;C组与D组间上述 3指标水平无显著性差异 (P >0 .0 5 )。光镜观察 ,B组退变较其余 3组明显 ;C组与D组间退变不明显 ;但 2组退变均甚于A组。表明痹康灵能抑制IL 1β、MMP 1、MMP 3的产生 ,具有抗炎、保护和修复关节软骨的作用 。
The objective of the paper is to observe the therapeutic effect of, and to explore the action mechanism of “BIKANG LING” (BKL, a TCD decoction for treating rheumatic or rheumatoid arthritis) on osteoarthritis (OA). Thirty-two Chinese white rabbits were divided into four groups (n=8 each): Group A not modeled as the normal control and Groups B, C and D OA-modeled with Hulth's method: Group B as the blank control, Group C as the Western medicine control and Group D as the TCD experimental. Three days postoperatively, Group A and Group B were given the normal saline; Group C and Group D were transgastrically given Rofexib solution and BKL (6 ml.kg-1/day), respectively. All animals were driven twice every day for 30 minutes since the first postoperative week; four weeks later, the articular cartilage and synovium were taken from the right tibial plateau and observed by optic microscope; the levels of IL (interleubin)-1β, MMP (matrix metalloproteinase)-1 and MMP-3 in the articular cartilage and synovium were determined by immunohistochemical method, respectively. The results showed that the levels of IL-1β, MMP-1 and MMP-3 of Group B were significantly higher than those of Group A (P < 0.01) and Group C (0.01<P < 0.05); so were the levels of IL-1β and MMP-3 of Group B than those of Group D (0.01<P < 0.05) and so was especially that of MMP-1 than that of Group D (P < 0.01); Groups C and D had no significant difference in the above three indices (P > 0.05); the optic microscopic observations showed: (1) severer degeneration in Group B compared with those in other three groups; (2) no significant difference between Groups C and D in the degeneration; (3) but severer degeneration of both Groups C and D than that of Group A, suggesting that BKL could inhibit the production of IL-1β, MMP-1 and MMP-3 and have the function of resisting the inflammation, protecting and repairing the articular cartilage, thereby producing some therapeutic effect on OA.
