摘要
目的 :观察骨肉瘤细胞对单核细胞侵袭能力的影响 ,探讨肿瘤浸润单核细胞协助肿瘤侵袭的作用 .方法 :利用transwell体外模型检测骨肉瘤细胞系MG6 3条件培养基作用下人单核细胞系Thp 1迁移及侵袭能力的变化 ;明胶酶谱方法检测MG6 3细胞条件培养基作用后Thp 1细胞表达MMP 9的改变 .结果 :与阴性对照组对比 ,MG6 3细胞条件培养基明显增强了Thp 1细胞迁移及侵袭能力 ;而且MG6 3细胞条件培养基对Thp 1细胞具有趋化作用 .明胶酶谱结果提示MG6 3细胞条件培养基作用后Thp 1细胞表达MMP 9增强 .结论 :骨肉瘤细胞MG6 3能够增强单核细胞Thp 1的迁移及侵袭能力 ,并可能是通过增强单核细胞MMP
AIM: To investigate the migration and invasiveness of monocytes co cultured with osteosarcoma cells. METHODS: An in vitro monocyte tumor c culture model was established, and the migration and invasiveness ability of human monocyte line Thp 1 after co culture was measured by Transwell test and the MMP 9 expression of monocytes was detected by gelatin zymography. RESULTS: Migration and invasiveness of Thp 1 cells after co culture were enhanced after co culture with osteosarcoma cells ( P <0.01) and gelatin zymography showed that the expression of MMP 9 in Thp 1 supernatant also increased. CONCLUSION: Osteosarcoma cells can alter the migration and invasiveness of monocytes, probably by the induction of MMP 9 expression.
