人膀胱癌细胞系BC-6的建立及其生物学特性

摘要 目的 :建立人膀胱癌细胞系BC 6 ,为膀胱癌的基础研究提供实验模型 .方法 :1 8例膀胱癌标本采用组织块直接培养法、8例采用细胞悬液培养法行原代培养 ;其中 1 2例同时行裸鼠皮下移植 ,在皮下生长后 ,连续裸鼠体内传代 3次 ,再行体外培养 .结果 :组织块直接培养和细胞悬液培养均未成功 .1 2例裸鼠皮下接种肿瘤有 3例生长 ,l例裸鼠意外死亡 ,l例裸鼠体内传代F2代肿瘤消失 .l例肿瘤裸鼠皮下接种后生长较快 ,裸鼠体内传代 3次后 ,取部分肿瘤组织体外培养得到膀胱癌细胞系BC 6 ,其形态结构 ,分化程度与原发瘤保持一致 ,染色体形态仍为人类核型 ,众数维持在 6 6~ 72之间 ,占82 % ,细胞周期分析G1 期 0 .5 8,G2 期 0 .0 8,S1 期 0 .35 ,细胞群体培增时间为 4 4h .结论 :通过裸鼠体内移植瘤建立的膀胱癌细胞系BC 6 ,与原发癌保持相同的生物学特性 ,体外连续传代1a以上 ,传代 92代 .细胞形态不变 ,生长周期恒定 。 AIM: To establish a new bladder cancer cell line and to provide a useful model for further research of bladder cancer. METHODS: Of the 26 specimens of human bladder cancer, 18 specimens were obtained by asepsis surgery and then cut into fragments and cultured, 8 specimens were homogenated to cell suspension and cultured and 12 specimens were implanted subcutanousely (SC) into nude mice. The transplanted tumor specimens grew SC and were passaged in nude mice for three generations and then the tumor tissues were cultured in vitro . RESULTS: Tissue culture and cell suspension culture failed to establish the bladder cancer cell line. Of the 12 subcutanousely implanted tumor specimens, three were found growing SC in three nude mice, respectively, but one of which died incidentally and the tumor disappeared in another after the second passage. Only one tumor grew SC at relatively more rapid speed, part of which, after three passages in the nude mice, was taken out and cultured in vitro , thus a bladder cancer cell line BC 6 was established. The established cell line resembled the original in tumor morphology and differentiation degree. The mode range of cell chromosome was 66-72. The cell cycle analysis showed that G 1 was 0.58, G 2 0.08 and S 1 0.35. The cell population doubling time was 44 h. CONCLUSION: A stable cell line has been established, for the bladder cancer cell line BC 6 is similar to the primary tumor in the biological characteristics and has been cultured in vitro continually for more than one year without characteristic changes.
