中药芪丹通脉片对动脉粥样硬化大鼠主动脉内皮细胞及PAI-1表达影响 被引量:7

摘要 目的 :探讨中药芪丹通脉片对动脉粥样硬化大鼠主动脉内皮细胞及PAI 1的影响 .方法 :将 72只SD雄性大鼠随机分为 6个实验组 ,其中一组为阴性对照组 ,给以普通饮食 ,一组为模型组 ,给予维生素D3,高脂和高胆固醇饮食 ,其余 4组在给予高脂、高胆固醇饮食建立动脉粥样硬化大鼠模型同时给予芪丹通脉片低剂量组 [0 .36g/ (kg·d) ]、中剂量组 [1 .0 8g/ (kg·d) ]、高剂量组 [3.2 4g/ (kg·d) ]、阳性对照新伐他丁组 [4mg/ (kg·d) ](n =1 2 ) .1 6wk后电镜观察大鼠主动脉内皮细胞损伤情况 ,并通过逆转录多聚酶链反应 (RT PCR)检测PAI 1表达 .结果 :模型组内皮细胞损伤最严重、PAI 1表达明显高于对照组 (P <0 .0 5 ) ;给予芪丹通脉片大鼠内皮细胞损伤减轻、PAI 1表达显著降低 ,中药高剂量组结果接近辛伐他丁组 .结论 :动脉粥样硬化大鼠主动脉内皮细胞损伤严重 ,其PAI 1表达增强 ,使用芪丹通脉片可明显改善内皮损伤程度 ,同时降低PAI 1表达 ,且其低、中、高浓度组PAI 1表达依次减少 (P <0 .0 5 ) . AIM: To explore the effect of Chinese herb QiDanTongMai Tablet (QDTMT) on the rat artic endothelial cell lesion and the expression of PAI 1 on aorta. METHODS: Seventy two male SD rats were divided into six experimental groups. Of the six groups, one was assigned randomly as the control group and was given normal diet ( n =12) and one as the model group and was given vitamin D3, high fat and cholesterol diet to make atherosclerosis models ( n =12). The other four groups, in addition to vitamin D3, high fat and cholesterol diet, were divided into low dose QDTMT group[0.36 g/(kg·d)], moderate dose QDTMT group[1.08 g/(kg·d)], high dose QDTMT group[3.24 g/(kg·d)] and Simvastatin group[4 mg/(kg·d)] for 16 weeks. The level of endothelial cell lesion was then observed by scanning electron microscope and the expression of PAI 1 on aorta in every group was detected by reverse transcript polymerse chain reaction (RT PCR). RESULTS: The level of the endothelial cell lesion and the level of PAI 1 expression in the model group were markedly higher than those of the control group. The significant decrease of PAI 1 expression was detected in groups taking QDTMT. The higher the dose of the QDTMT, the lower the expression of PAI 1. CONCLUSION: QDTMT can decrease the level of endothelial cell lesion and the expression of PAI 1 on aorta in a dose dependent manner in rats with atherosclerosis.
