蜕皮甾酮对Aβ_(25-35)细胞毒的保护作用

The protective effects of ecdysterone on the PC12 cells induced by Aβ_(25-35)

目的 观察蜕皮甾酮 (Ecdysterone ,ECR)对Aβ2 5 3 5诱导的PC12细胞毒的保护作用。方法 PC12细胞受损及存活情况分别由乳酸脱氢酶释放量 (LDH)及MTT法检测。结果 10 0 μmol·L 1Aβ2 5 3 5与PC12细胞作用2 4h时 ,MTT吸光值减小 ,LDH释放量则明显增加 (均 P <0 .0 1)。如果用 1 10 0 μmol·L 1ECR预处理PC12细胞0 .5h ,则Aβ2 5 3 5的PC12细胞毒作用明显被抑制 (ECR5 0 ,P <0 .0 5 ;ECR10 0 ,P <0 .0 1)。结论 ECR能够抑制Aβ2 5 3 5的毒性 。

Objective To study the effect of Ecdysterone (ECR) on the PC12 cell cytotoxicity induced by beta-amyloid peptide fragment 25-35(Aβ 25-35 ). Methods PC12 cell survial was monitored by LDH release and 3-(4,5- dimethylthiazol-yl)-2,5-diphenyltetrazolium bromide (MTT) assays. Results Treatment of PC12 cells with Aβ 25-35 (100μmol·L -1 ) for 24 h caused a great decrease of MTT absorbance as well as the increase of LDH activities from PC12 cells ( all P<0.01 ). After the cells had been pretreated with 1-100 μmol·L -1 ECR for 0.5 h, ECR attenuated the cytotoxic effect of Aβ 25-35 (ECR50, P<0.05; ECR100, P<0.01). Conclusions The cytotoxicity of PC12 cells induced by Aβ 25-35 , could be inhibited by ECR.
