羊膜抑制兔角膜成纤维细胞凋亡及其TGF-β_1mRNA表达的实验研究

摘要 目的 探讨羊膜移植抑制眼表纤维组织增生的机制。方法 将传代的兔角膜成纤维细胞接种于羊膜基质面 ,分别培养 1和 7d ,采用吖啶橙法观察羊膜对成纤维细胞凋亡的影响。同时采用RT PCR技术 ,观察成纤维细胞中TGF β1mRNA表达的变化。结果羊膜对培养 1和 7d的兔角膜成纤维细胞均有明显地抑制凋亡作用。同时发现在羊膜基质面培养 1d的兔角膜成纤维细胞 ,其TGF β1mRNA的表达与对照组相比无明显改变 ,而培养 7d后 ,TGF β1mRNA的表达明显下降。结论 羊膜有抑制体外培养的兔角膜成纤维细胞凋亡的作用。同时还有抑制体外培养的兔角膜成纤维细胞TGF β1mRNA表达的作用 ,可部分解释羊膜抑制纤维增生的机制 ,为临床应用提供理论依据。 Objective To investigate the mechanism of the inhibitation of ocular fibrosis by amniotic membrane transplantation. Methods After the primary culture of the rabbits' corneal fibroblasts,they were seeded on amniotic membrane matrix for 1 day or 7 days. Fibroblasts apoptosis was studied by acridine orange. TGF-β_1 mRNA in rabbits,corneal fibroblasts were examinee through creverse transcription-polymerase chain reaction.Results Amniotic membrane inhibited the fibroblasts apoptosis significantly which were seeded on amniotic membrane matrix for 1 day or 7 days. At the same time ,TGF-β_1 mRNA in rabbits,corneal fibroblasts which were seeded on amniotic membrane matrix for 1 day were not changed,but they were suppressed significantly when the fibroblasts were cultured on amniotic membrane for 7 days. Conclusion Fibroblasts apoptosis can be inhibited and TGF-β_1mRNA in rabbits'corneal fibroblasts in vitro can be suppressed by amniotic membrane matrix ,which can partially explain the effect of amniotic membrane on inhibitation of fibrosis.
