摘要
目的 探讨水分离时应用 5 氟尿嘧啶 (5 FU)对兔后发性白内障形成的影响及其对兔眼的毒性作用。方法 4 0只兔随机分为对照组与用药组 ,在兔晶状体囊外摘出术中 ,用 0 .3mL浓度分别为 0 .0 5、0 .1、0 .2 g·L-1的 5 FU在晶状体囊袋内进行水分离 ,使其直接短暂作用于晶状体上皮细胞。术后观察 3个月 ,行后囊混浊评分及照相 ,比较各组晶状体后囊混浊情况 ;光镜观察晶状体上皮细胞增殖情况并评价药物的毒性作用。结果术后 3月对照组后囊混浊评分 5只为 3,2只为 2 ,高于用药组 (1只为 1) ,差异有显著性(P <0 .0 1)。用药组后囊混浊的发生率低于对照组 ,差异有显著性 (P <0 .0 5 )。组织病理学检查显示 ,对照组的晶状体上皮细胞增殖较用药组明显 ,各组虹膜及睫状体均未有慢性炎症反应情况出现。结论 水分离时使用 5 FU的给药方式更为快捷方便 ,可能会成为一种更好的防治后发障的临床应用方案。
Objective To investigate whether 5-FU can inhibit posterior capsule opacification (PCO) without causing ocular toxicity.Methods Forty rabbits were randomly divided into control and 5-FU groups. Different concentrations of 5-FU(0.05,0.1,0.2g·L -1 ) were injected into capsular bags by hydrodissection during ECCE in rabbit eyes. The changes of the cornea,anterior chamber and posterior capsule were observed by slit-lamp microscope,ophthalmoscope and photography,and the toxicity of 5-FU were evaluated.Results The PCO score and PCO rate of the control group were higher than those of the 5-FU group(P<0.05).Histological analysis showed that the proliferative activity of the control group was much more intense than that of the 5-FU group,The iris and ciliary body in each group got no chronic inflammatory.Conclusion The results suggest that the application of 5-FU by hydrodissection effectively reduce PCO in rabbit eyes,which may be more suitable for the prevention of PCO.
