摘要
目的 确定可溶性支架转反义 PCNA是否具有抑制静脉移植物内膜增殖的效果。方法 2 5只健康纯种新西兰大耳白兔随机分为对照组 ,支架组 ,正义组 ,反义组 ,错配组。将颈外静脉移植于同侧颈总动脉后用可溶性支架将反义 ,正义及错配的基因片段导入静脉移植物。术后 2 8d进行表达检测。结果 (1)反义组静脉桥的内膜厚度为(4 2 .72± 3.792 )μm,显著低于对照组、支架组、正义组及错配组 (79.0 0± 1.2 2 5 )μm ,(75 .0 4± 3.2 4 2 )μm ,(74 .77±8.5 35 )μm ,(78.6 1± 7.816 )μm,P<0 .0 1。 (2 )反义组内膜与中膜比值为 0 .6 8± 0 .2 12 ,明显低于对照组、支架组、正义组及错配组 (1.32± 0 .0 6 6 ,1.35± 0 .198,1.33± 0 .2 5 8,1.39± 0 .2 0 0 ,P<0 .0 1)。结论 运用可溶性支架转反义PCNA基因可明显抑制内膜的增殖。
Objectives To evaluate whether intimal hyperplasia can be inhibited through transcatheter delivery of PCNA ODNs with soluble stents. Methods Twenty-five New Zealand rabbits were randomized into five groups(five in each).After models were constructed, sense, antisense, mismatch oligonucleotides were administered locally through soluble stents.Anmials were killed at 28 days and defined segments of vein grafts were sectioned and observed undered microscope. Resultsthe intimal thickness in PCNA antisense ODN groups significantly decreased compared with control,stent,sense, mismatch groups P<0.01.The ratios of intimal to medial thickness for PCNA antisense ODN groups significantly decreased compared with control,stent,sense, mismatch groups P<0.01. ConclusionIntimal hyperplasia of rabbit vein graft can be markedly suppressed by intravascular local delivery of synthetic antisense PCNA via soluble stent.
