摘要
目的 :观察红花对实验性大鼠肝纤维化的影响 ,并探讨其作用机制。方法 :在 CCl4 诱导实验性大鼠肝纤维化 3周后 ,给予红花 5 .0 g/ kg,每天 1次腹腔注射 ,同时设正常对照及 CCl4 对照组。分别用放免法检测血清透明质酸 (HA)、层粘连蛋白 (L N)、3型前胶原 (PC )。HE及 VG法染色观察组织的病理变化。免疫组织化学法检测肝组织 Desmin的表达。结果 :红花治疗组大鼠肝组织纤维化程度计分、血清 HA、L N、PC 浓度、肝组织中 Desmin染色阳性细胞面积低于 CCl4 对照组 ,差异显著。结论 :红花抑制肝星状细胞的激活和转化 ,具有一定的抗肝纤维化作用。
Objective: To investigate the inhibitive effects of safflower on experimental hepatic fibrosis in rats and its mechanism.Methods: After the rats were treated with CCl 4 for 3 weeks to induce the experimental rat liver fibrosis,the intraperitoneal injection was performed in rats with safflower at the dose of 5.0g/kg ,once daily.Therefore we formed the safflower treatment group and the CCl 4 control group:The serum levels of hyaluronic acid(HA),laminin(LN) and pro-collagen type Ⅲ peptide (PCⅢ) were detected by radioimmunoassay.Hepatio fibrosis scoring grade was assessed through Hematoxilin-Eosin and Van-Gieson staining and observed under light microscope..Expression level of Desmin was determined by immunohistochemica staining method.Results: In the treatment group,the liver fibrosis scores, serum levels of HA,LN ,PCⅢand Desmin positive cell proportion in the liver were significantly lower than those of CCl 4 control group.Conclusion: safflower can inhibit hepatic stellate cells from transforming into myofibroblasts and has a certain effect on prohibition if hepatic fibrosis.
