摘要
本文观察了六种生物膜稳定剂抑制石英对肺泡巨噬细胞膜的破坏作用。将家兔麻醉,无菌操作,行肺灌洗获取健康家兔的肺泡巨噬细胞。将细胞配制成10×10~5cells/ml悬液,置于CO_2孵育箱中培养。与临床治疗矽肺的药物(矽肺宁)作对比。通过测定细胞内外酸性磷酸酶(ACPase)含量变化,筛选五种试剂对膜具有最大稳定的浓度。分析结果表明,膜保护作用由强到弱的排列顺序是Zn^(++),Se,矽肺宁,VE,Mg^(++),Ca^(++)。本文结果提示,石英直接损伤肺泡巨噬细胞的生物膜,故任何能保护生物膜的试剂,对煤工矽肺的防治均可能有意义。
The inhibitory effects of six kinds of biomembrane stabilizer to the destructive action by quartz on biomembranes of alveolar macrophage were observed.Broncho-alveolar lavage fluid was taken from anaesthetized animal. Alveolar macrophages were suspended in RPMI-1640 medium as 1.0×105 cells/ml, then cultured.Five kinds of biomembrane stabilizers as well as the clinical drug.Xi Fei Ning (Zn++,Se ,VE ,Mg+ + , Ca+ + ) were analysed by detecting , the conte.nt changes of acid phosphatase (ACPase) . The concentrations of stabilizers were selected at first.The results indicated that membrane protective actions of the six agents in series from strong to weak were Zn++, Se, Xi Fei Ning, VE ,Mg+ + ,Ca++. The results showed that silicon dioxide may directly damage the biomembrane of alveolar macrophage.So the biomembrane stabilizers may be significant in the treatment of silicosis.
