二甲基苯蒽诱发家兔卵巢肿瘤及其生物学特性的初步观察 被引量:2

摘要 目的 以二甲基苯蒽 (DMBA)原位埋线法诱发家兔卵巢肿瘤 ,并初步观察其生物学特性。方法 棉线浸入加热熔化的DMBA中 ,取雌性家兔 5只 ,分别在其双侧卵巢埋入含有DMBA的棉线 ,连续观察 8个月 ,处死荷瘤家兔 ,通过大体、光镜、电镜以及流式细胞技术等方面进行肿瘤生物学特性的初步观察。结果  1只家兔因术后感染而死亡 ,2只观察 8个月无肿瘤发生。 2只分别发生未分化癌及腺癌 ,组织学特点与相应的人卵巢肿瘤很相似 ;流式细胞分析S期细胞含量分别为 4 0 .72 %和2 9 .87%。结论 二甲基苯蒽能原位诱发较大型动物家兔的卵巢肿瘤 。 Objective To establish an ovarian tumor model in rabbits with 7,12-dimethylbenz[a]anthracene(DMBA) and observe its biological features. Methods DMBA was heated, where in the central portion of a cotton yarn was immersed in it. The DMBA-coated yarn was inserted into the ovaries of 5 rabbits. The animals sacrificed after 8 months were observaed. Studies were proceed through microscope, electron microscope(EMS), and flow cytometer (FCM). Results One rabbit was died of severe infection after operation, 2 rabbits were still tumor-free after 8 months, while 2 were formed undifferentiated carcinoma and adenocarcinoma,respectively. It is quite similar with ovarian tumor of woman in biological features. The S-phase of each tumor was 40.72% and 29.87% separately. Conclusion DMBA can induce ovarian tumor in rabbits and it can serve as a big animal model for the study of ovarian tumor.
