摘要 目的 观察二氯化钴 (CoCl2 )诱发缺氧对胰腺癌PC 3细胞系核因子 κB(NF κB)表达的影响。方法 采用免疫组织化学方法检测NF κB在PC 3细胞系中的表达 ,并通过逆转录 聚合酶链反应 (RT PCR)观测CoCl2 与NF κB表达间的量 效和时 效关系。结果 胰腺癌PC 3细胞系中存在NF κB的表达 ,CoCl2 可上调细胞中NF κB的表达 ,并且其和NF κB表达间体现出一定剂量和时间范围内的依赖性关系。结论 胰腺癌PC 3细胞系中存在着NF κB的表达 ,通过CoCl2 诱导缺氧可以上调NF Objective To investigate the effect of CoCl 2 on the expression of NF-κB in pancreatic carcinoma PC -3 cell line by inducing hypoxia. Methods The expression of NF-κB in pancreatic carcinoma PC-3 cell lines was studied by immunohistochemistry. The time-concentration relationship and time-effect relationship between CoCl 2 and NF-κB were explored by reverse transcription polymerase chain reaction (RT-PCR). Results NF-κB was expressed in pancreatic carcinoma PC-3 cell line. CoCl 2 up-regulates the expression of NF-κB in PC-3 cell line in a time-dependent and dose-dependent manner. Conclusion NF-κB was expressed in PC-3 cell line, and CoCl 2 can up-regulate the expression of NF-κB in pancreatic carcinoma PC-3 cell line by inducing hypoxia.
