全反式维甲酸对人食管癌细胞增殖及DNA聚合酶β表达的影响 被引量:4

摘要 目的 研究全反式维甲酸对人食管癌细胞诱导分化作用。方法 在体外细胞培养的基础上 ,观察细胞形态学的变化 ,细胞增殖动力学的变化。采用MTT比色法测定细胞增殖抑制率 ,免疫组化和westernblot方法检测DNA聚合酶 β(polβ)蛋白表达。结果 经全反式维甲酸处理后 ,人食管癌Eca10 9细胞的细胞形态趋向良性分化 ,细胞增殖指数明显降低。polβ表达降低。结论 全反式维甲酸对人食管癌Eca10 9细胞有诱导分化作用 ,其机理可能是抑制 polβ的表达 ,改变细胞的形态结构和生物学特性 。 Objective To ditermine the effect of all-trans retinoic acid(ATRA) on human esophageal cell. Methods The Eca109 esophageal cells were exposed to all-trans retinoic acid. The morphological and functional changes of the cells were observed with MTT. The DNA polymerase β expression was assayed by immunohistochemistry and western blot. Results After Eca109 cells were treated with all-trans retinoic acid for 5d, the malignant phenotypes of the cells gradually change to the muture cells, the proliferation index decreased markedly. The expression of the pol β protein increased dramatically. Conclusion All-trans retinoic acid can induce the human esophageal cancer cell differentiation, the mechanism of differentiation may be inhibition of the expression of morphological and structural characteristic of the cancer cells and induction of the differentiation.
