端粒酶调控蛋白(hTRT)在膀胱癌组织表达情况研究

摘要 目的 研究膀胱癌组织中端粒酶hTRT蛋白表达情况。方法 应用免疫组化方法对 5 2例膀胱癌组织和 30例膀胱正常组织进行hTRT蛋白表达研究 ,其中 30例膀胱癌组织和 30例膀胱正常组织同时进行hTRT蛋白westernblot检测。结果 免疫组化检测显示 4 9例膀胱癌组织的hTRT蛋白表达阳性 ( 94 % ) ,对照组正常膀胱粘膜阳性表达的只有 1例 ( 3.4 % )。两者有明显差异 (P <0 .0 1) ,肿瘤分级与肿瘤组织hTRT蛋白表达情况无相关性。westernblot检测结果中膀胱癌组织阳性表达为2 5例 ( 83% ) ,正常膀胱组织粘膜均为阴性结果。结论 膀胱癌组织hTRT蛋白有明确的阳性表达 ,其表达情况与肿瘤的分级无相关性 ;而正常膀胱粘膜则无hTRT蛋白表达。膀胱癌组织的hTRT蛋白与端粒酶活性表达可能相关。 Objective To study the expression of hTRT protein in bladder tumor tissues. Methods The expression of hTRT protein in 52 bladder tumor tissues and 30 bladder normal samples were estimated by immunohistemical techniques. Other 30 tumor tissues and 30 normal tissues were detected by western blotting and TRAP methods. Results Forry nine tumor samples show positive expression of hTRT(94%), while only 1 normal samples show positive reaction(3.4%). The expression level of hTRT protein of bladder cancer was also significantly different from normal tissues( P <0.01). Twenty five bladder cancer cases express hTRT, while no normal tissues had positive expression of hTRT. The expression level of hTRT of bladder cancer was significantly different from normal bladder( P <0.01).Conclusion Bladder cancer tissues have increased positive expression of hTRT protein, yet the expression has no relationship with cancer grades. The expression of hTRT protein may correlate with Telomerase activity.
