摘要
目的 测定肺癌组织中 p5 3抑癌基因突变蛋白的表达 ,诱生型一氧化氮合酶 (iNOS)的表达及分布。了解肺癌组织中p5 3抑癌基因突变蛋白与iNOS的关系。方法 按自身配对原则收集人肺癌癌灶组织 ,癌灶周围组织及远癌组织各 4 0份 ,分别进行免疫组化法测定 p5 3抑癌基因突变蛋白的表达 ,原位杂交法测定iNOSmRNA的表达及分布。 结果 1、肺癌组织中p5 3抑癌基因突变蛋白的表达阳性。与肺癌的临床分期、组织分型、淋巴结转移未见相关 ,(均为P >0 .0 5 )。 2、iNOSmRNA在肺腺癌中的表达高于鳞癌 ,(均为P <0 .0 5 )。 3、肺腺癌组织中iNOS在 p5 3抑癌基因突变蛋白表达阳性组高于p5 3阴性组 ,(P <0 .0 1)。结论 肺腺癌中 p5 3抑癌基因突变蛋白阳性组iNOS表达高于p5 3阴性组的表达 ,提示NO/iNOS途径可诱使p5
Objective To explore and evaluate the expression of p53 gene mutation protein and the expression of induced nitric oxide syntheses (iNOS) in tumor tissue of non-small cell lung cancer,and the relation between them.Methods Ever 40 samples of human lung cancer,paracancerous and distant cancerous tissues with self-pair control were collected.HE staining and immunohistochemical SP method were used to observe the expression of p53 and the iNOS expression was determined by nucleic acid in site hybridization.Results 1.The mutation of p53 tumor suppressor gene in lung cancer,showed no correlation with clinical stage,histology and lymphnode metastases( P >0.05).2. iNOSmRNA expression in lung cancer tissue was significantly higher than in paracancerous and distant cancerous tissue.Among them,there is no difference,( P >0.05).The iNOS mRNA expression in human lung adenocarcinoma samples was significantly higher than that in lung squamous carcinoma( P <0.05).3.The expression of iNOS in p5 3 mutation-positive samples is significantly higher than that in p53 mutation-negative samples in lung adenocarcinoma,( P <0.01).Conclusion The expression of iNOS in p53 mutation-positive samples is significantly higher than that in p53mutation-negative samples in lung adenocarcinoma,witch may demonstrated NO/iNOS pathway can induce p53 to mutate,( P < 0.01 ).
