胸痹通胶囊对高脂血症大鼠肝低密度脂蛋白受体基因表达的影响

Effect of Xiongbitong Capsule on Gene Expression of LDL Receptor in Rats

目的 在基因水平上探讨中药胸痹通胶囊的降脂作用机制 ,为中药新药的研制提供科学依据。方法 将 40只大鼠随机分为正常对照组、高脂对照组、胸痹通胶囊实验观察组、血脂康胶囊药物对照组 ,每组 10只。正常对照组给予正常饲料喂养 ;高脂对照组饲以高脂饲料 ;胸痹通胶囊组给予高脂饲料加胸痹通胶囊 12 g/(kg·d) ;血脂康对照组给予高脂饲料加血脂康 0 .2 16g/(kg·d)。连续 3 0d后 ,RT -PCR法测定大鼠肝低密度脂蛋白受体信使核糖核酸 (LDLRmRNA)水平。结果 与高脂对照组比较 ,胸痹通胶囊组能明显提高高脂血症大鼠肝LDLRmRNA水平 (P <0 .0 1) ;与血脂康组比较作用无统计学意义(P >0 .0 5 )。结论 胸痹通胶囊可明显增强高脂血症大鼠肝LDLR基因表达 ,从而加快血中低密度脂蛋白胆固醇(LDL -C)的清除 ,起到调控血脂水平的作用 ,该作用与血脂康相仿。

Objective To investigate the mechanism of Xiongbitong capsule in decreasing serum lipid in gene level.Methods 40 wistar rats were divided into 4 groups randomly including rats of normal group fed with normal diet,rats of high lipid group fed with high fat diet,rats of Xiongbitong capsule group fed with high fat diet and Xiongbitong capsule 12 g/(kg·d),Xuezhikang group fed with high fat diet and Xuezhikang 0.216 g/(kg·d).After 30 days,the levels of livel LDLR mRNA were detected by RT-PCR.Results Compared with the higt fat group,the Xiongbitong group can significantly increase the gene expression of LDLR mRNA in liver (P<0.01).But there was no significant different between the Xiongbitong group and Xuezhikang group(P>0.05).Conclusion The effect of Xiongbitong capsule regulationg serum lipid might associate with its increasing the gene expression of liver LDLR and increasing the excretion of serum LDL-C.This effect is similar to Xuezhikang.
