鳖甲煎丸对免疫性肝纤维化大鼠TGF-β_1mRNA表达的影响 被引量:14

摘要 目的 :观察中药复方鳖甲煎丸对免疫性纤维化模型大鼠肝组织细胞因子TGF -β1 mRNA表达的影响 ,分析鳖甲煎丸的治疗作用 ,探讨鳖甲煎丸抗肝纤维化的作用机理 ;并与西药秋水仙碱作对照 ,为临床防治肝纤维化用药提供科学依据。方法 :雄性Wistar大鼠 90只随机分为 6组 :正常对照组、模型对照组、秋水仙碱预防组、秋水仙碱治疗组、鳖甲煎丸预防组、鳖甲煎丸治疗组 ,选用猪血清诱导免疫损伤性肝纤维化模型。各组于 10周后取 10只大鼠断头处死 ,取肝组织 ,用原位杂交法检测肝组织内TGF -β1 mRNA。结果 :鳖甲煎丸能够明显减少TGF -β1 mRNA的表达 ,作用接近或优于秋水仙碱 ,显示它具有明显的抗纤维化作用。鳖甲煎丸和秋水仙碱的预防效果同治疗效果相比差异均不显著。 Objective:To study the effect of BIEJIAJIANWAN(BJJW),a combination of traditional Chinese medical herbs,on TGF-β 1mRNA in rat hepatic tissue.Combining with pathology and configuration change of rat liver,we can analyze curative effect of BJJW and discuss it's mechanism in anti-fibrosis.It is a kind of further research on Zhang Zhongjing's prescriptions and herbs with the view of molecular biology.Methods:Ninety Wistar rats(male,180~200g weight) were divided randomly into six groups: blank contrast group,model contrast group,BJJW prevention group,BJJW treatment group,Colchicin prevention group and Colchicin treatment group.15 rats per group.Pig's serum was choosen to beinducement of immunity damnification hepatic fibrosis model.Testing TGF-β 1mRNA by in situ hybridization.Results:Transcribe of TGF-β 1mRNA in the immunity hepatic fibrosis rat liver.BJJW has remarkable anti-fibrosis ablity.It can also prevent collagen synthesis and motivate the degradation of collagen fiber.Emergence of TGF-β 1mRNA are clearly restrained by BJJW in the experiment.It indicates that its anti-fibrosis effect may be realized by restraining these cytokines.The cytokines emergence can't be restrained by Colchicin.It indicates that Colchicin dosen't possess these mechanisms.There isn't significant difference between the effect of prevention and treatment with using two medicines.
