摘要
研究以腹腔注射博莱霉素A5复制肺纤维化模型 ,观察肺纤煎对关键性致肺纤维化的细胞因子TNF -αmRNA表达及蛋白分泌的影响 ,旨在为进一步探讨本法抗纤维化的作用机制。目的 :观察肺纤煎对博莱霉素致小鼠肺纤维化的干预作用及机制。方法 :以腹腔注射博莱霉素A5复制肺纤维化模型 ,将小鼠随机分组并给予不同药物治疗 ,从病理改变、肿瘤坏死因子 (TNF) -αmRNA表达及蛋白分泌等不同方面 ,动态评价肺纤煎对肺纤维化的治疗作用。结果 :肺纤煎治疗后肺泡炎、肺纤维化明显减轻 ;TNF -αmRNA表达及蛋白分泌均有不同程度的降低。结论 :肺纤煎可通过调控TNF -αmRNA表达及蛋白分泌而减轻博莱霉素A5致小鼠肺纤维化。TNF -α具有多种生物活性 ,不但可以引起和加重炎症反应 ,而且对成纤维细胞有促生长活性。有报道证实 ,在体内TNF -α局部释放可以引起局灶性成纤维细胞和胶原的聚集 ,在动物和肺间质纤维化患者支气管肺泡灌洗液中 ,肺泡巨噬细胞分泌的TNF -α较正常对照组增加 ;动物实验发现用抗TNF -α单克隆抗体可减轻肺间质胶原的沉积 ,再次肌注重组TNF -α ,胶原沉积又明显增加 ,提示TNF -α在肺纤维化的发生。
Objective:To evaluate the therapeutic effects and mechanism of FeiXianJian in bleomycinA-5(BLMA-5)-induced pulmonary fibrosis in mice.Methods:We established the anmoral model of pulmonary fibrosis induced by bleomycin administered intraperitoneally,divided mices into different groups and gave different treats,evaluated it with the pathologic changes,the level of activity TNF-α and expression of messenger RNA for TNF-α to investigate the effect of FeiXianJian with prednisone dynamically.Results:The results indicated that the alveclitis and fibrosis alleviated significantly,the activity of TNF-α and expression of messenger RNA for TNF-α after FeiXianJian treatmeant.Conclusion:We supposed FeiXianJian could regulate the activity of TNF-α and expression of messenger RNA for TNF-α so that it could alleviate the process of BLMA-5-induced pulmonary in mice.
