摘要 目的 探讨 β淀粉样蛋白 (Aβ)对大鼠神经干细胞凋亡诱导作用。 方法 分离、培养胚胎大鼠神经干细胞 ,Aβ 2 5~ 35 (2 0 μmol/L)与神经干细胞共育 4 8h后 ,原位检测神经干细胞凋亡。 结果 Aβ 2 5~ 35作用后细胞死亡率上升 ,4 8h后死亡率趋于平稳 ,凋亡检测表明细胞主要通过凋亡途径死亡。结论 Aβ 2 5~ Objective To explore the apoptosis induction of amyloid peptide on neural stem cells in embryonic rats.Methods Rat′s embryonic neural stem cells in vitro were isolated and cultured, then these cells were treated with amyloid peptide Aβ 25-35 (20 μmol/L)for 48 hours and the apoptosis of neural stem cells were investigated with TUNEL technique.Results The death rate of neural stem cells raised after treatment with Aβ 25-35 and amounted to the peak at the 48th hours. Further research showed that apoptosis was the main form of death.Conclusion Aβ 25-35 can induce neural stem cells to apoptosis.
