摘要
目的 :研究链脲佐菌素 (STZ)诱导的糖尿病 (DM)大鼠血管舒缩活性改变与一氧化氮 (NO)、前列环素 (PGI2 )、内皮超极化因子 (EDHF)等的关系。观察人参白虎汤复合活性部位 (RB)的保护作用。方法 :采用STZ(6 0mg/kg ,ip)造糖尿病大鼠模型 ,合格动物进行分组 ,分别每日用RB 10 0 ,5 0mg/kgig及格列本脲 (Gly 10mg/kg·d-1ig)干预治疗。 75d后分别记录在L 精氨酸 (L Arg) ,L 硝基精氨酸 (L NOArgNLA)及吲哚美辛 (Ind)存在(或不存在 )下大鼠胸主动脉环对苯肾上腺素 (Phe )及乙酰胆碱 (Ach)引起的收缩及舒张反应。结果 :Phe引起的最大收缩 ,DM组 (1 2 2± 0 0 8g)高于正常组 (0 86± 0 0 8g) (P <0 0 5 )。NLA阻断NO合成后 ,Phe引起的最大收缩增加 ,增加部分反应NO基础释放。DM组Phe引起的最大收缩增加 (0 0 2± 0 0 2g)低于正常组 (0 2 5± 0 0 3g)(P <0 0 1) ,说明DM组NO基础释放较正常组减少 92 %。L Arg对抗Phe收缩活性 ,模型组及正常组Phe收缩分别减少 (0 0 4± 0 0 2 3g)、(0 2 9± 0 0 7g) (P <0 0 5 ) ,减少部分反应NO的最大释放 ,与正常组相比DM组NO的最大释放减少 86 %。与正常组 (4 9 37± 5 95 ) %相比 ,Ach在模型组引起的舒张效应明显下降 (2 6 6 3± 5 93) % (P<0 0 5 )。
AIM:To study the change of aorta contraction and relaxation induced by nitric oxide (NO),prostacyclin(PGI 2)and endothelium-drived hyperpolarizing factor(EDHF)in diabetic rats induced by STZ and the protective effect of combined active parts from Ginseng Baihu Decoction (RB). METHOD: Diabetic rats were Induced by streptozotocin(STZ) (60 mg/kg ig).After 75 days of treatment,the responses to phenylephrine (Phe),and acetylcholine (Ach) on isolated rat aorta rings were recorded and compared in the absence and presence of L-argine(L-Arg),N-Nitro-L-argine(NLA) and Indomethacin (Ind). RESULT:Phe induced maximum contraction was reduced in diabetic model (1.22±0.08 g) vs normal group (0.86±0.08 g)(P<0.05).By blocking NO synthesis with NLA,maximum contraction induced by Phe in model group decreased (0.02±0.02 g) vs (0.25±0.03 g)in normal group(P<0.01).This showed that the basal NO release decreased in model group vs normal group by 92%. L-Arg decreased Phe contracting activity in model group(0.04±0.03) g vs(0.29±0.07) g(P<0.05)normal group,the NO maximum release decreased by 86 % in model group vs in normal group. Ach induced relaxation was reduced in model group (26.63±5.93%) vs normal group (49.35±5.65%)(P<0.05).Relaxation induced by PGI 2 & EDHF was also reduced in model group. The combined active parts from Ginseng Baihu Decoction can improve vascular impairment. CONCLUSION:Macrovascular contraction and relaxation of diabetic rats were abnormal. RB can improve them and be used to treat vascular complications.
