摘要
研究了空心菜优良变异植株嫩茎愈伤组织的诱导、分化、试管苗生根、移栽、扦插及田间栽培所需要的条件,建立起优良变异空心菜嫩茎的无性系及快速繁殖技术.结果证明,诱导嫩茎愈伤组织的理想培养基是MS+BA0.8mg/L+2,4 D0.3mg/L;诱导愈伤组织分化的理想培养基是MS+La(NO3)30.8mg/L;生根的理想培养基是1/2MS+IAA0.2mg/L;以炉灰渣为试管苗的移栽扦插基质,移栽成活率为100%,扦插成活率为98.6%.试管苗田间栽培试验还证明,这种试管苗生长旺盛,可以收获二次,产量提高了约一倍.
The conditions were studied for induction, diferentation, radication, transplatation, and graftage of the callus of Lpomoea aquetica and its regenerative clone was established. The best medium for callus induction is MS+BA0.8 mg/L+2,4-D0.3 mg/L. The high differentiation frequencies obtained in MS+La(NO_3)_3mg/L 1/2MS+IAA0.2 mg/L is suitable for radication. With cinder as medium, a survival ration of 100% for transplatation and that of 98.6% for graftage were observed. This tube spout grew well, reaped twice and the yield was doubled.
