兔骨髓间充质干细胞分离方法的比较

Comparison of different methods for mesenchymal stem cell isolation from rabbit bone marrow

目的 建立并优化骨髓间充质干细胞分离纯化的方法。方法 从兔的胫骨中抽取骨髓 ,分别通过直接培养、经Ficoll-Hypaque和Percol两种分离介质进行分离后再培养的方法 ,收获所培养的细胞 ,然后测量间充质干细胞所占的比例。结果 直接培养、经Ficoll-Hypaque、Percol分离后再培养的细胞中 ,骨髓间充质干细胞比例分别为6 7.3%、83.6 %、93.4 %。结论 采用Percol分离方法可以更好的提高MSCs的纯度 。

Objective To evaluate the efficiency of two different methods of mesenchymal stem cell isolation from rabbit marrow. Methods The rabbit bone marrow was taken out and treated with the following three methods: ①to culture these cells directly; ②to isolate the cells with Ficoll-Hypaqueor before culture; ③ to isolate the cells with Percol before culture. The harvested cells were cultured and measured to find out the proportion of mesenchymal stem cells. Results The proportion of mesenchymal stem cells is diffient: Group 1 is 67.3%, Group 2 is 83.6% and Group 3 is 93.4%. Conclusion Application of Percol to isolate cells from rabbit bone marrow before culture is the most effective way to get mesenchymal stem cells.
