摘要
目的 :探讨白细胞介素 1β对人牙髓细胞金属蛋白酶 1(matrixmetalloproteinase 1,MMP 1)、环氧合酶 2 (cyclooxygenase 2 ,COX2 )基因表达变化的生物学意义。方法 :培养人牙髓细胞 ,传代至第 4代 ,PT PCR检测人牙髓细胞是否表达IL 1β受体 ,进而用 1nmol/L人的重组IL 1β刺激人牙髓细胞 18h ,提取细胞总RNA ,反转录为cDNA ,半定量PCR检测IL 1β对人牙髓细胞的金属蛋白酶 1、COX2的mRNA的表达变化。结果 :人牙髓细胞表达IL 1β受体 ,而且用 1nmol/L人的重组IL 1β明显地刺激牙髓细胞的金属蛋白酶 1、COX2的mRNA表达。结论 :牙髓炎时牙髓组织内的IL 1β生成增多 ,会进一步引发金属蛋白酶 1、COX2基因异常表达相应增多 ,而金属蛋白酶 1导致炎症基质降解、COX2将引发炎性痛的病理改变。
Objective:To study the effect of endogenous IL-1β on the expression of matrix metalloproteinases (MMPs) and COX2 in dental pulp cells. Methods:Human dental pulp cells were treated with human recombinant IL-1β at 1 nmol/L in serum-free medium for 18 h. Then the cells were collected and total RNA was isolated, MMPs and COX2 mRNA expression was assessed by semiquantitative RT-PCR. Results:IL-1β at 1 nmol/L induced the expression of COX2 and MMP-1 mRNA in human dental pulp cells. Conclusion:IL-1β may contribute to stimulating expression of MMPs and COX2 in the dental pulp during pulpitis.
