摘要 目的 建立菟丝子中槲皮素的含量测定方法。方法 采用高效液相色谱-紫外检测法(HPLC-UV),以卡马西平为内标,菟丝子灌胃给药后,大鼠含药血清样品经液-液萃取后在C18反相色谱柱上进行分离测定。结果 槲皮素血药浓度在0.05~9 μg/mL范围内线性关系良好(r2=0.9996)。方法回收率为86.65%~99.07%,提取回收率为79.91%~84.56%,最低检测限为0.05 μg/mL。结论 本方法稳定可靠,结果可为菟丝子在临床用药的安全性和有效性提供依据。 Objective To find the method of assaying quercetin level in dodder seed. Methods The quick and sensitive HPLC-UV method was established with carbamazepine acting as internal standard. After the rats were douched with dodder seed, drug-containing serum samples of rats were separated and measured on C18 reverse-phase chromatography column after being liquid to liquid extracted. Results The linear range of calibration curve is 0.05~9 靏/mL (r2=0.9996). The method showed good precision and recoveries for serum with 86.65%~99.07%, extracting recoveries 79.91%~84.56%, and the limit of detection (LOD) is 0.05 靏/mL. Conclusion The method is stable and reliable, and the results can provide scientific basis of safety and effectiveness for clinical use of dodder seed.
