两系创伤反应明显差异小鼠肺血管通透性及Scyb15基因基础表达量的比较研究 被引量:1

摘要 目的 通过对两系创伤反应明显差异小鼠伤前、伤后肺血管通透性的及生理状态下Scyb15基因的比较研究 ,初步探讨创伤反应差异的相关解剖结构基础 ,为创伤相关基因背景的研究提供实验和理论依据。方法 以对创伤反应有明显差异的两系近交系小鼠为研究对象 ,采用BST Ⅰ型生物激波管致伤模型 ,利用荧光分光光度计测定肺组织在伤前伤后荧光素钠的含量 ,以此定量反映肺血管通透性的变化 ;采用荧光定量PCR技术 ,对Scyb15基因在生理状态下的表达进行定量。结果 两系小鼠间比较 ,在伤后 1h及伤后 4h ,肺血管通透性有显著性差异 (P <0 .0 5 ) ;同系小鼠伤前和伤后比较 ,BALB c在伤后 3 0min ,其血管通透性已显示出明显差异 (P <0 .0 5 ) ,而C5 7BL 6在伤后 1h才显示出差异 (P <0 .0 5 ) ;不管是伤前伤后 ,在同一时间点 ,BALB c小鼠的血管通透性均有较C5 7BL 6增高的趋势 ;伤后 4h内 ,两系小鼠的血管通透性呈现持续升高 ;生理状态下 ,Scyb15基因在C5 7BL 6小鼠中的表达量较在BALB c小鼠中的表达量低 ,且二者相比有显著性差异 (P <0 .0 5 )。结论 BALB c小鼠和C5 7BL 6小鼠间Scyb15基因基础状态下及创伤后肺通透性变化的显著性不同可能是创伤反应差异的重要原因 ,这两系小鼠可能存在先天性 (遗传性 )肺血管结构上? Objective To explore the physiological structure base of heterogeneity of traumatic responses by means of comparison of lung microvascular permeability and expression of Scyb15 (small inducible cytokine B subfamily, member 15) in physiological state between BALB/c and C57BL/6 mice. Methods BALB/c (BG) and C57BL/6 (CG) mice were employed in this study. After the establishment of the animal injury model using BST Ⅰ Bioshock Tube, the concentrations of fluorescein sodium (FINa) in the lungs in each group were measured with flurospectrophotometer respectively before injury and at 0.5, 1, and 4 h after injury. The expression of Scyb15 in physiological state in each group was measured by quantitative fluorescent PCR. The results from the two groups were compared by using Student, t test. Results At 1 and 4 h after injury, the level of FINa in BG was higher than that in CG. Compared with that in CG, the distinct change of the level of FINa in BG was earlier; the level of Scyb15 in BG was higher than that in CG. There was significant difference between BG and CG ( P <0.05). Conclusion The difference in microvascular permeability and Scyb15 expression between BALB/c and C57BL/6 mice may possibly be the reason for heterogeneity of traumatic responses, which might be resulted from the difference in the physiological structure base of lung microvascular permeability in the two lines of mice.
