摘要
目的 :研究三苯氧胺 (tamoxifen ,TAM)对大鼠子宫平滑肌细胞增殖的影响及其可能的机制。方法 :采用细胞培养、细胞计数、MTT、流式细胞技术和激光共聚焦显微镜技术。结果 :TAM(10 -6mol·L-1)使大鼠子宫平滑肌细胞的生长曲线上移。TAM(10 -8~ 10 -6mol·L-1)剂量依赖性的促进大鼠子宫平滑肌细胞的增殖作用。TAM(10 -6mol·L-1)使大鼠子宫平滑肌细胞周期由G1期加速向S期转化 ,G1期的DNA含量由对照组的 5 5 .5 %下降到加药组的 32 .8% ,S期的DNA含量由对照组的 2 9.0 %上升到加药组的 4 9.4 %。激光共聚焦检测到TAM(10 -6mol·L-1)可使大鼠子宫平滑肌细胞内的Ca2 + 浓度显著升高。结论 :TAM可以通过调节细胞周期各阶段DNA含量和细胞内Ca2 + 浓度水平 ,从而调节大鼠子宫平滑肌细胞的增殖活动。
Objective:To investigate whether tamoxifen is involved in the proliferation of the cultured rat cervical muscle cells and explore its possible mechanism.Methods:The level of cell proliferation was estimated by drawing proliferation curves of rat cervical muscle cells,assessing OD value by MTT assay,techniques of flow cytometry and laser scanning confocal microscope. Results: Tamoxifen (10 -6 mol·L -1 ) shifted the curve upward. Tamoxifen (10 -8 to 10 -6 mol·L -1 ) significantly stimulated the level of cell proliferation. Tamoxifen (10 -6 mol·L -1 ) stimulated G_1/S transfer of the cell cycle and increased the releasing of calcium obviously. Conclusion:It is suggested that tamoxifen modulates the proliferation of the cultured rat cervical muscle cells. Tamoxifen may stimulate the proliferation through modulating cell cycle and calcium in the cells.
