摘要
通过RT PCR和3′ RACE方法,分别从鲇鱼(Silurusasotus)和斜带石斑鱼(Epinepheluscoioides)中克隆了核糖体大亚基蛋白L15(ribosomalproteinL15,RPL15)的完整cDNA,并详细分析了其序列特征.这两种鱼的RPL15蛋白cDNA均包含一个615个核苷酸的完整阅读框,推测分别编码一个204个氨基酸的蛋白.不论是核苷酸序列还是推测的氨基酸序列,这2个序列分别与同属的南方大口鲇和同科的大眼鳜rpl15序列同源性最高,与其他已知的脊椎动物rpl15序列也有较高的同源性.基于这2个序列的编码区和其他鱼类的rpl15编码序列进行分子系统学分析,与传统的形态分类结果是一致的.
The full-length cDNAs of ribosomal protein L15 (RPL15) were cloned by RT-PCR and (3′-RACE) respectively from Silurus asotus and Epinephelus coioides. The two cDNAs individually include an ORF of 615 bp which encodes a protein of 204 amino acid. Alignment analysis indicated that both the deduced amino acid sequences and the nucleotide ones of the two cDNAs showed high identity to other known vertebrate rpl15 sequences, with the highest identity between the sequences from Silurus asotus and Silurus meridionalis, Epinephelus coioides and Siniperca kneri. In addition, the two ORF sequences were used to construct phylogenetic trees with other piscine rpl15 ORFs, and the topologies of the obtained trees were largely congruent with the classical systematics.
