摘要
利用MetaMorph软件定量分析了中国莲(NelumbonuciferaGaertn.)每条前中期染色体上(从短臂到长臂)二脒基苯基吲哚(DAPI)荧光强度的变化,结合染色体的相对长度和臂比作为辅助参数,构建了中国莲前中期染色体的定量染色体图.该定量染色体图是实际的前中期染色体的一种直观模式,可识别中国莲基因组中每条染色体以及分析染色体上的异染色质分布,为莲的细胞遗传学研究提供了一种新的方法.
Prometaphase chromosomes of Nelumbo nucifera Gaertn.(2n=16)which were longer than metaphase chromosomes were quantitatively analyzed based on their distribution pattern of DAPI staining patterns. The DAPI staining patterns showed prominent uneven distribution along the chromosomes. Based on the DAPI staining patterns (from short arm to long arm), with chromosomal relative length and arm ratio as supplementary parameters, the quantitative chromosome map was constructed for the first time in Nelumbo nucifera Gaertn. by the MetaMorph software. The quantitative chromosome map was the intuitional mode of actual premetaphase chromosomes. It could not only identify every chromosome in genome but also analyze fractional area of heterochromatin in chromosomes. The quantitative chromosome map is therefore considered to be a new method for cytological studies of lotus.
