摘要
以大豆分离蛋白为原料,选用4种商业蛋白酶水解获得大豆蛋白抗高血压活性肽。以水解度为指标,设计正交实验,确定了最佳的水解条件,利用紫外分光光度法测定酶在最佳的水解条件下,随水解时间不同所得酶解产物对血管紧张素转换酶的抑制效果。最终得到As1.398中性蛋白酶,在pH值为8.0,温度为45℃,E/S(酶与底物浓度比)6%,S(底物浓度)4%时,水解480min所得水解产物对ACE(血管紧张素转换酶)的抑制效果最强,IC50为0.846g/L。
In this study,we choice four trade-Enzymes to hydrolyze Soybean Protein Isolate for obtaining active-peptides.We design orthogonal experiment to find the best conditions of hydrolyze for aiming at Degree of Hydrolyzing.Through using ultra-violet spectrophotometer,we have found out that have different inhibitory effect through different hydrolysis time,which have different hydrolysis production.We have found Enzyme A is the best Enzyme,at the optimum conditions of hydrolysis pH=8.0,45 ℃,E/S 6%,S 4%;through hydrolyzing 480 min,its inhibitory value of IC50 is 0.846 g/L to Angiotensin Converting Enzyme.
