甾体激素对乳腺癌细胞钠碘转运体基因表达的调控 被引量:2

摘要 目的通过甾体激素对乳腺癌细胞钠碘转运体(NIS)基因表达的影响,探讨乳腺肿瘤组织NIS的表达特点和调控因素。方法乳腺癌细胞(MCF7)进行培养,分别给予不同浓度的甾体激素包括雌二醇、孕酮、睾酮和地塞米松等刺激72h。采用RT鄄PCR方法检测细胞中NIS的mRNA表达情况。结果乳腺癌细胞MCF7在经过雌二醇、孕酮和睾酮刺激后,细胞中NIS的mRNA表达比对照组有明显增加,并且与刺激物浓度成正相关。较大剂量地塞米松则明显抑制乳腺癌细胞中NIS的mRNA表达。结论甾体激素对乳腺癌细胞NIS基因表达具有调节作用。 Objective: To investigate the effect of steroid hormone(Estradiol, Progesterone, Testosterone and Dexamethasone)on gene expression of sodium-iodide symporter(NIS) in breast cancer cell(MCF7). Methods: The breast cancer cell was cultured with negative control culture or culture with different concentrations of steroid hormone for 72 h. NIS gene mRNA in breast cancer cells cultured was determined by reverse transcriptase-polymerase chain reaction(RT-PCR). Results: Gene mRNA of sodium-iodide symporter can be found increasing along with the concentration of Estradiol, Progesterone and Testosterone dose-dependently, but decreasing after administration of Dexamethasone. Conclusion: Steroid hormone may play a role in iodide-uptake modulating in breast cancer cells by their effect on NIS gene expression.
