摘要
目的:将血管内皮生长因子受体3胞外第二免疫球蛋白区(VEGFR-3 2-Ig)的反义基因在体外进行原核表达,并将产物进行生物活性鉴定。方法:构建VEGFR-3 2-Ig反义基因的原核表达载体,经过原核表达、亲和层析及蛋白酶切后获得VEGFR-3 2-Ig的反义多肽,用MTT法进行活性鉴定。结果:获得VEGFR-3 2-Ig的反义多肽,MTT法证明VEGFR-3 2-Ig的反义多肽具有较强生物学活性(细胞增殖抑制率可达68.7%)。结论:通过原核表达成功获得VEGFR-3的反义多肽,为研究和利用反义多肽提供了实验理论依据。
Objective: To obtain the prokaryotic expression of VEGFR-3 antisense in vitro and identified the activity of the VEGFR-3 antisense peptide. Methods: Construct the prokaryotic expression plasmid of VEGFR-3 antisense, then acquire the VEGFR-3 antisense peptide through the prokaryotic expression, purification and PreScission cutting, and identified the activity of antisense peptide by MTT. Results: VEGFR-3 antisense peptide was acguired and proved to be bio-active by MTT. Conclusions: Obtaining the VEGFR-3 antisense peptide through the prokaryotic expression successfully would be a base for the research and use of antisense peptide.
