摘要
目的 研究苯并 [a]芘 (BaP)、高温及其联合作用对小鼠脑组织热应激蛋白 70 (HSP70 )与热应激蛋白 90 β (HSP90 β)表达的影响。方法 将 5 6只昆明种小鼠随机分为 7组 (每组 8只 ) ,即空白对照组、溶剂对照组、低剂量BaP单独染毒组、高剂量BaP单独染毒组、单独高温处理组、高温处理 +低剂量BaP染毒组、高温处理 +高剂量BaP染毒组。低、高剂量BaP染毒分别给予BaP 0 5mg/kg和 5mg/kg ,用植物油作溶剂 ,每周 4次腹腔注射 ,溶剂对照组用植物油作平行处理 ,空白对照组不做处理。高温处理为每周4次 ,每次 4 1 5~ 4 2 0℃处理 2h。实验 8周后取各组小鼠脑组织制作混合匀浆 ,Westernblot法检测HSP70和HSP 90 β水平。 结果 HSP70在高温单独作用、低剂量BaP单独作用、高温与低剂量BaP联合作用下表达明显升高。HSP90 β仅在高温与低剂量BaP联合作用及高温与高剂量BaP联合作用下表达升高。 结论 不同的热应激蛋白在不同条件下对高温。
Objective To study the effects of benzopyrene(BaP)or/and heat on levels of heat stress proteins 70(HSP70) and heat stress proteins 90β(HSP90 β)in mice brain.Methods 56 mice were randomly divided into seven groups,i e ,standard control group,vehicle control group,low dose BaP group,high dose BaP group,heat group,heat+low dose BaP group,and heat+high dose BaP group.After 8 experiment weeks,the mice were sacrificed;their brains were removed and made into homogenates.Detect levels of HSP70 and HSP90 β in homogenates by the method of western blot.Results For HSP70 level,in the low dose BaP group,heat group and heat+low dose BaP group,HSP70 level was much higher than that of control groups.For HSP90 β level,in the heat+low dose BaP group and heat+high dose BaP group, HSP90 β level was higher than that of control groups.Conclusions Different HSPs under different conditions play preventive and marking roles on damaged brain tissues induced by hazards such as heat and BaP.
