摘要
目的建立完善军团菌分离培养的检验方法,提高阳性检出率。方法用分离培养法,依据菌落形态和在缺乏L-半胱氨酸的平板和血平板不生长初判军团菌,再用血清凝集进行分型,结合生化反应和PCR结果进行确证。结果用本文方法成功检测出军团菌,在2003年对广州地区各种水体进行军团菌检测,121宗水样中分离出军团菌31宗,总污染率为25.6%,其中公共场所冷却塔水中军团菌污染率为43.48%。阳性率与国内外报道接近。结论本方法有很好的分离培养效果,广州地区部分水体中存在军团菌污染,尤以公共场所冷却塔水中军团菌污染情况较为严重。
Objective To establish and improve methods of isolation and cultiv ation of Legionella and hence to enhance the positive detection rate. Method Aft er isolation and culture, Legionella colonies were preliminarily distinguished o n blood plate(negative) and plate without L-cysteine. The bacteria was confirme d by serotyping and PCR. Results Legionella was successfully detected employing the method in this study. 31 Legionella strains were identified. The rate of env ironmental contamination was 25.6%(31/121), especially in public air conditione r cooling water with a rate of 43.48%. The positive detection rate of Legionell a was comparable with that reported overseas. Conclusions The method used was ef fective in isolating and culturing Legionella. Legionella contaminations were fo und in some water bodies in Guangzhou, especially the public water-cooling towe rs.
