高含量大豆磷脂酰胆碱的提纯工艺研究 被引量:6

摘要 研究了以浓缩大豆磷脂为原料,采用丙酮和乙醇作溶剂,经丙酮脱油、真空干燥、乙醇抽提、减压蒸馏浓缩制备高含量磷脂酰胆碱(PC)的提纯工艺。丙酮和乙醇可回收,反复使用。在丙酮用量200mL、萃取时间15min、萃取温度40℃的最佳脱油工艺条件下得到了丙酮不溶物含量为97.6%的中间产物———粉末磷脂。乙醇提取试验选用L1645正交表进行,考察了乙醇浓度、固液比、温度、时间、抽提次数对PC含量的影响。结果表明:影响卵磷脂含量的主要因素是温度和乙醇浓度,其最佳条件为固液比1∶20、乙醇浓度95%、抽提时间30min、温度35℃、抽提次数2次,PC含量富集到45.1%,提取率可达到62.0%。 In order to improve the content of phosphatidylcholine(PC), a study has been carried out on the use of acetone and alcohol to extract PC from soybean lecithin. After degreased with acetone, dried in vacuum, extracted with alcohol and decompressed condense at last, the purer phosphatidylcholine(PC) was prepared. Acetone and alcohol could reclaim. A result showed that acetone insoluble residue was up to 97.6% with acetone volume 200mL, extraction time 15 min and temperature 40℃. The experiment of alcohol extraction adopted the L_(16)4~5 orthogonal test and inspected the effect of alcohol concentration, soybean lecithin/ alcohol, extraction time, temperature and times to content of PC. It was found that the main influencing factors of PC content were temperature and alcohol concentration. The best conditions obtained by the orthogonal tests were as follows: soybean lecithin∶alcohol 10∶200, alcohol concentration 95%, extraction time 30 min, temperature 35℃, extract twice,The PC content and yield were 45.1% and 62.0% respectively.
