再生障碍性贫血T细胞早期激活及激活潜能的研究 被引量:1

Early activation and activated potential of T lymphocytesin patients with aplastic anemia

摘要 目的:研究再生障碍性贫血穴AA雪患者外周血CD4+、CD8+T淋巴细胞培养前后早期激活标志CD69的表达及其意义。方法:将外周血在植物血凝素(PHA)20μg/ml条件下进行全血细胞培养,于0h和4h分别用双色免疫荧光标记流式细胞仪对CD4+、CD8+T淋巴细胞CD69的表达进行分析。结果:PHA刺激前SAA患者CD4+、CD8+细胞CD69的表达率分别为(8.96±7.23)%和(10.67±7.58)%,CAA患者CD8+细胞CD69的表达率为(7.36±5.49)%;PHA刺激后AA患者CD4+、CD8+细胞表达CD69明显增强,SAA为(71.73±11.91)%和穴61.74±13.44雪%鸦CAA为(59.35±10.15)%和(48.78±8.25)%熏CD4+细胞CD69的表达率高于CD8+细胞。结论:T细胞早期活化及其活化潜能增强,产生针对自身造血干/祖细胞的细胞毒效应在AA发病中起重要作用。 Objective: To investigate the expression of T cell early activation marker(CD69) on CD4+ and CD8+ lymphocytes in peripheral blood of patients with aplastic anemia (AA) and the pathophysiological significance of the changes in CD69 expression in different T cell populations of both pre-and post-culture in vitro. Methods: The whole blood cell culture procedure was applied to activate T lymphocytes by PHA (20 μg/ml) in vitro. The expression rates of CD69 of CD4+ and CD8+ lymphocytes 0 h and 4 h after culture were analyzed by two-color flow cytometry. Results: The expression rates of CD69 of CD4+ and CD8+ cells in patients with SAA were(8.968)% respectively, and the expression rate of CD69 of CD8+ cells in patients with CAA was(7.3648.78or role in the pathogenesis of AA.
