摘要 目的:探讨CASPc光敏物质数字血管造影在脉络膜黑色素瘤中浓聚的动态变化,以寻找最佳光动力治疗时间。方法:TopconImageNetH1024成像系统和TopconTRC鄄50鄄VT眼底照相机连接,使用CASPc特制的光滤器(吸收峰在580nm,发射峰695nm)于兔耳静脉注射CASPc5mg/kg进行CASPc数字血管造影成像。12只带瘤动物在注射CASPc后1、24、48、72h分别成像。8只动物与血管造影成像同一时间眼球摘除行荧光显微镜检查。将二组结果进行光敏物质肿瘤最大浓聚比较。结果:数字血管造影成像显示最大荧光发生在注射后48h。荧光光度结果也证明最大光敏物质浓聚在注射后48h。结论:本研究提示,CASPc血管造影成像是决定光动力治疗最佳时间的有效方法。 Objective: To examine chloroaluminum sulfonated phthalocyanine(CASPc) as a modality for detecting dye accumulation in the experimental choroidal melanomas, and to optimize the treatment time for photodynamic therapy(PDT) of occular melanoma. Methods: Digital angiograms were performed using a Topcon ImageNet H1024 imaging system, a KODAK Megaplus camera, and a Topcon-50-VT fundus camera. Filters were specifically designed for chloroaluminum sulfonated phthalocyanine (CASPc) (peak absorption at 580 nm and peak emission at 695 nm). The concurrent images were obtained when CASPc(5 mg/kg) was injected via ear veins of 12 tumor-bearing rabbits, and the follow-up images 1, 24, 48 and 72 hours after the injection. Fluorescence microscopy was performed in the other 8 rabbits. The angiograms were taken when the tumor-bearing eyes were enucleated at the same time points of angiograms. The maximum dye accumulations of the two sets were compared. Results: Digital images showed that maximal fluorescence occurred 48 hours after injection. Fluorescence photometry results also demonstrated maximum dye accumulation 48 hours after the injection. Conclusion: This study suggests that the CASPc angiography is a valid modality for determining the treatment time of PDT.
