小鼠睾丸初级精母细胞染色体畸变试验方法改进及应用

Method improvement for chromosome aberration test of mouse primary spermatocyte

目的 对小鼠睾丸初级精母细胞染色体畸变试验方法进行改进 ,以达到染色体分裂相增多、铺展好、背景清晰的效果利于阅片 ,并将改进后方法应用于农药的致突变实验。方法 将原方法中分离曲细精管改为全睾剪碎 ,软化步骤提前 ,并改用 2 %柠檬酸钠软化 ;固定过程中去除大块组织 ,最终制备小鼠睾丸初级精母细胞染色体标本。结果 两次实验中 ,某农药各剂量组染色体畸变细胞率与阴性对照组比较差异无显著性 (P >0 0 5) ;某农药各剂量组及阴性对照组染色体畸变细胞率与阳性对照组比较差异有显著性 (P <0 0 1)。结论 方法改进后可见大量分散铺展良好、背景清晰中期分裂相细胞 ,利于观察。

ObjectiveTo improve the methods of chromos om e aberration test of mouse primary spermatocyte. Methods Enhan cing the intenerate, scissoring the spermary and useing 2% sodium citrate to int enerate, and finally making chromosome slice. ResultsThere was n't significant difference between the negative controlled and the treated group s ( P >0 05),while significant difference was showed between the positive c ontrolled and the above two groups ( P <0 01). ConclusionImproved methods had more mitotic figure to be obse rved.
