摘要
近年来,放射肿瘤学研究者致力于采用特异性阻断剂阻断DNA修复过程中关键调控基因,并通过有效抑制DNA损伤修复,促进肿瘤细胞死亡,收到较好的肿瘤放疗效果。但是同时发现,阻断剂技术本身存在诸多缺陷,这极大地限制其尽快应用于临床肿瘤治疗。自小片段干扰核糖核酸(smallinterferenceRNA,siRNA)干涉技术发现以来,其强大的阻断真核细胞中特异表达蛋白的能力为众多研究者所关注。更重要的是,它较好地解决了应用阻断剂带来的上述问题,从而为肿瘤放射基因治疗研究提供了新途径。
Recently, many radiooncology researchers had gained some good curative effects on DNA damage repair inhibited and tumor cell death increased by blocking key control genes in the course of DNA repair with specific blocking agents. Otherwise, the blocking agents have many defects, which extremely lim-ited its application on curing tumor. Since small interference RNA (siRNA) technology was discovered, its powerful function of blocking expressible specific protein in eukaryocytes was attented by many researchers. Meanwhile, this technology admirably resolved mentioned above problems caused by using the blocking agents, which provided a new way for tumor radiogene therapy.
