摘要
目的 :检测P73、细胞周期素 (Cylin)B1蛋白在皮肤血管瘤组织中的表达 ,探讨P73、CylinB1在血管瘤发生发展中的作用及临床意义。方法 :采用免疫组织化学SP法检测增生期血管瘤、退化期血管瘤、正常皮肤组织 (各2 0例 )中P73、CylinB1蛋白的表达 ,并采用病理图像分析系统定量分析和比较P73、CylinB1蛋白在增生期血管瘤、退化期血管瘤和正常皮肤组织中的表达。结果 :在增生期血管瘤、退化期血管瘤和正常皮肤组 ,P73蛋白免疫组化阳性反应颗粒的平均光密度分别为 :6 .4 0 8± 2 .15 1,1.0 73± 0 .5 16 ,0 .95 3± 0 .12 0 ;阳性面积率分别为 :0 .184± 0 .0 15 ,0 .0 98± 0 .0 14 ,0 .0 87± 0 .0 12 ;CylinB1蛋白免疫组化阳性反应颗粒的平均光密度分别为 :0 .14 6± 0 .0 16 ,0 .0 79± 0 .0 18,0 .0 6 8± 0 .0 2 1;阳性面积率分别为 :0 .5 38± 0 .0 6 5 ,0 .2 0 4± 0 .0 79,0 .176± 0 .0 5 4。增生期血管瘤与退化期血管瘤、正常皮肤组分别相比 ,P73、CylinB1蛋白阳性表达的平均光密度和阳性面积率差异均有极显著性 (P <0 .0 0 1) ,消退期血管瘤与正常皮肤组之间 ,P73、CylinB1蛋白阳性表达的差异均无显著性 (P >0 .0 5 )。结论 :P73、CylinB1蛋白在血管瘤发生发展和毛细血管内皮细胞的异常过度增生中?
Objective: To investigate the quantitative expressions of P73 and Cyclin B 1 protein and their roles in the carcinogenesis and development of children hemangioma. Methods: The quantitative expressions of P73 and Cyclin B 1 protein in 40 hemangioma and 20 normal skin tissues were detected by immunohistochemistry and image analysis. Results: In proliferative hemangioma and involutional stage of hemangioma and normal skin tissues, average optical density of P73 protein positive immunostaining was 6.408±2.151, 1.073±0.516 and 0.953±0.120 respectively, and positive area rate was 0.184±0.015, 0.098±0.014 and 0.087±0.012, while average optical density and positive area rate of Cyclin B 1 protein positive immunostaining was 0.146± 0.016 ,0.079± 0.018 ,0.068±0.021, and 0.538±0.065,0.204±0.079,0.176±0.054 respectively. Compared with those in involutional stage of hemangioma or normal skin tissues, there were significant increases of P73 and Cyclin B 1 protein positive expression in proliferative stage of hemangioma (P<0.001 ). No statistical significances of P73 or Cyclin B 1 expressions were observed between involutional stage of hemangioma and normal skin tissues (P>0.05). Conclusion: P73 and Cyclin B 1 may play a role in the carcinogenesis and development of children hemangioma.
