硝化池中氨氧化细菌amoA基因的检测及其多样性研究 被引量:5

摘要 为了分析污水处理系统中氨氧化细菌的种群组成 ,筛选合成了一对对氨氧化细菌氨单加氧酶基因 (amo A )特异结合的引物序列 ,利用 PCR技术对从活性污泥中抽提的细菌总 DNA进行扩增 ,得到不同重组子的 amo A序列片段 .运用 BL AST程序将测序结果与基因库中的公开序列进行比较 ,发现在该污水处理系统中分布有大量亚硝化单胞菌属 (N itrosomonas)细菌 ,其中最主要的是欧洲亚硝化单胞菌 (N itrosomonas europaea) ,由此推测N In order to analyze the composition of ammonia-oxidizing communities in the wastewater treatment system, a pair of primers special for the amoA gene was designed and used to amplify the specific DNA fragment from the total bacterial DNA extracted from the wastewater. After TA cloning and DNA sequencing, different sequences were compared with the available databases by using the BLAST network service. A large quantity of Nitrosomonas genus which is mostly Nitrosomonas europaea was found in the wastewater treatment system. The result suggested that Nitrosomonas genus probably played a predominant role in the ammonia oxidation in the system.
