摘要
目的 探讨胆固醇合成抑制剂洛伐他汀 (Lovastatin ,LOV)对HL 6 0细胞凋亡的影响及其可能的分子机制。方法 采用光镜、透射电镜、DNA梯形带检测以及流式细胞仪分析等技术方法 ,观察LOV对HL 6 0细胞形态、超微结构和凋亡的影响 ,以及Fas抗原表达的改变。结果 LOV处理HL 6 0细胞 2d后 ,光镜下细胞数量明显减少 ,细胞形态发生不同程度的改变 ;透射电镜下 ,HL 6 0细胞呈现凋亡性变 ;有凋亡特征性的DNA梯形带出现 ;Fas抗原的平均荧光强度增加。结论 LOV不仅能抑制HL 6 0细胞增殖 ,而且可诱导HL 6 0细胞凋亡 ;LOV能上调HL 6 0细胞Fas抗原表达 ,可能是LOV抑制HL 6 0细胞增殖并诱导其凋亡的重要分子机制。
Objective To study the effects of lovastatin (LOV) on the apoptosis of HL-60 cells and its molecular mechanisms. Me- thods The morphological and ultrastructural changes in LOV-treated HL-60 cells were observed using light microscope and electron microscope. The changes of apoptosis and Fas antigen expression in LOV-treated HL-60 cells were detected by DNA ladder assay and flow cytome- try (FCM), respectively. Results The morphology and number of HL-60 cells were obviously changed under the light microscope after treatment with LOV for 2 d. The apoptotic change was detected with the electron microscope and DNA ladder. The expression of Fas antigen was increased significantly in LOV-treated HL60 cells. Conclusion LOV not only can inhibit the cellular proliferation of HL-60 cells but also can induce the apoptosis of HL-60 cells, in which LOV effects of up-regulating Fas antigen expression in HL-60 cells may play a key role.
