湖北保康不同花色野生紫薇的RAPD分析 被引量:18

摘要 紫薇为千屈菜科紫薇属木本花卉,作为一种观赏、药用兼环保的优良花木,在现代城市园林绿化中有着广泛的应用前景,湖北保康是全国乃至全世界紫薇的一个重要发祥地。湖北保康野生紫薇种质资源非常丰富,由于长期的天然杂交、自然变异,形成了丰富多彩的自然类型。本研究基于RAPD分析技术,利用筛选出的12个多态随机引物,对4种不同花色的湖北保康野生紫薇样品进行了扩增反应,共得到41个多态位点。利用POPGENE软件包对RAPD数据进行分析,结果表明保康野生紫薇具有较丰富的遗传多样性,4种不同花色群体间遗传多样性参数存在较大差异,紫色群体的遗传多样性高于其它三个群体。利用UPG-MA方法对不同花色样品间的聚类分析结果表明红色、白色及粉色系的单株基本聚在一起,但紫色系的单株聚类结果比较分散。根据不同花色群体间的遗传距离对4个群体进行聚类,结果表明紫色与红色的亲缘关系较近,与粉色的亲缘关系较远。该研究通过对不同花色野生紫薇遗传多样性的分析,有利于在DNA分子水平上揭示不同花色品种间差异、亲缘关系及相互之间遗传物质的交流情况,这为紫薇花色遗传改良提供了有利的理论依据。同时该研究对有效保存和合理利用保康野生紫薇资源具有重要意义。 Lagerstroemia indica is a kind of woody flower which belongs to Lythraceae family Lagerstroemia genus. Lagerstroemia indica has been extensively applied to gardens virescence in modern city as a kind of plant with the use of view and admire, drug and environment protection. The Baokang count in Hubei province is one of cradles of Lagerstroemia indica in china even in world. Wild Lagerstroemia indica resource from Hubei Baokang is very much abundant, and exists in rich and colorful natural type formed by natural hybridization and natural variation of long-term, The amplification of Lagerstroemia indica in 4 different colors from baokang county in hubei province has been done and 41 polymorphic loci has been obtained based on 12 polymorphic random primers screened by RAPD analysis. The results which been analyzed using POPGENE software show that relatively rich genetic diversity exists in wild population of Lagerstroemia indica from Baokang and extinct difference of genetic parameters exists in 4 populations in different colors. Based on UPGMA method, the results of cluster analysis of samples in different colors indicate that the samples of red, white and pink gather unanimous basically, but the samples of purple gather relatively scattered. The result of cluster analysis of populations in different colors show that purple population and red population has relatively near relation, relatively far relation between purple and pink in terms of genetic distance. The study is of benefit for revealing difference, relationship and exchange among varieties in different colors from the level of DNA molecule and provides advantageous theory foundation theoretical foundation for color improvement of Lagerstroemia indica. And the study is important for effective conservation and reasonable usage of wild Lagerstroemia resource from Baokang.
