摘要
①目的 研究海洋贝类提取物 (EMS)抗血小板聚集及抗氧化作用 ,探讨其抗动脉粥样硬化 (AS)作用机制。②方法 采用二磷酸腺苷 (ADP)和胶原作诱导剂进行体内及体外血小板聚集实验 ,观察EMS对大鼠血小板聚集功能的影响。采用快速微量法检测EMS对血清总超氧化物歧化酶 (T SOD)及脂质过氧化物丙二醛 (MDA)含量的影响。③结果 EMS体外用药 (9.76、2 9.2 7、4 8.78g/L)可显著降低胶原诱导的家兔血小板聚集 (F =1 2 .6 1~ 6 7.2 0 ,q =2 .76~ 1 1 .37,P <0 .0 5、0 .0 1 ) ;在体内实验中 ,EMS(5、1 0、2 0 g/kg)灌胃给药对ADP诱导的大鼠血小板聚集率无明显影响 (F =0 .39~ 2 .0 1 ,P >0 .0 5 ) ;EMS(5、1 0、2 0g/kg)灌胃给药可显著升高大鼠血清T SOD的活性 (F =85 .4 3,q=9.2 0~ 1 9.6 1 ,P <0 .0 1 ) ,EMS 1 0 g/kg灌胃并可降低MDA含量 (F =9.6 8,q =3.6 1 ,P <0 .0 5 )。
Objective To study the anti oxidation and inhibitory action on platelet aggregation of the extract from marine shellfish (EMS) and its mechanism of anti atherosclerosis. Methods The platelet aggregation was induced by adenosine diphosphate (ADP) and collagen in vivo and vitro. The serum superoxide dismutase(SOD) and malondiadehyde (MDA) were measured with microcontent fast method. Results EMS (9.76,29.27,48.78 g/L) in vitro significantly depressed the collagen induced platelet aggregation ( F=12.61-67.20, q=2.76-11.37, P <0.05, 0.01); EMS(5,10,20 g/kg) had no effect on the ADP in vivo induced platelet aggregation( F=0.39-2.01,P >0.05);EMS(5,10,20 g/kg)evidently raised serum T SOD( F=85.43,q= 9.20- 19.61,P <0.01); and only 10 g/kg reduced serum MDA in rats( F=9.68,q=3.61,P <0.05). Conclusion EMS can inhibit platelet aggregation and promote anti oxidation.
